IRON, CALCIUM ANTIOXIDANT WITH CYANOCOBALAMIN, MULTIVITAMIN & MULTIMINERAL SYRUP

IRON, CALCIUM ANTIOXIDANT WITH CYANOCOBALAMIN, MULTIVITAMIN & MULTIMINERAL SYRUP is a multi-component range designed to combine ingredients such as iron, calcium, cyanocobalamin, vitamins, minerals and antioxidants in a controlled syrup system. As this range contains several different categories of micronutrients, the formulation development requires a systematic approach to ingredient compatibility, mineral dispersion, pH, viscosity, colour, bland flavour balance and microbiological stability. This formulation can be developed for pharmaceutical customers looking to include a nutritional liquid range in their PCD Pharma Franchise range with quality documentation and packaging.

Formulation development of Iron-Calcium Combinations in a Stable Vehicle

A detailed evaluation of single ingredients and their physicochemical and functional behaviour in the target liquid vehicle will take place, and the formulation design will be adjusted accordingly.

The liquid vehicle formulation will contain purified water with sweetening agents, viscosity modifiers, stabilizers, buffers, flavouring ingredients, antimicrobial agents and other functional excipients where required.

Iron and calcium compounds may affect the final composition and require specific formulation development attention as the target salts can affect pH, appearance, solubility, colour, clarity and potential precipitation in combination with all of the other nutrients. The formulation for this range will also need to be evaluated for ingredient compatibility and mineral stability when together in the liquid phase.

The cyanocobalamin and other vitamin ingredients in this range can also have specific handling sensitivities requiring further formulation of the liquid carrier formulation. This approach can provide a suitable environment for maintaining ingredient integrity.

Dispersion of Multiple Minerals in a Stable Syrup

A multi-mineral syrup will need more than simple mixing of individual components and may require suspension of certain particulates to maintain liquid homogeneity. The particle size, density and wetting properties of certain ingredients will need to be evaluated in formulation development trials and sources of agglomeration or sedimentation may need to be minimised.

Wetting agents, suspending agents or stabilisers can be incorporated as required. The manufacturing step sequence will need to be established according to the solubility profile of individual ingredients, and certain components can be pre-mixed into the main batch where appropriate.

The bulk can be stirred at a controlled speed for a designated period while recording pH, colour, stability parameters such as clouding or flocculation and sediment formation. Samples can be drawn from different locations within the vessel and analysed for homogeneity.

Colour and flavour Control in an Iron and Calcium-Containing Range

pH control can be critical in formulations containing a mixture of mineral salts and water-soluble vitamins. The pH range can affect the solubility, presence of sediments, colour, antioxidant and stabilizer effectiveness, and the overall appearance of the formulation.

The pH range required can be established during formulation development based on the complete composition rather than individual ingredients. Buffers can also be used as needed to maintain formulation stability during storage.

A known colour specification can be applied to formulations containing a number of active minerals and vitamins. Changes in colour can be monitored during formulation development and stability testing. Appearance of the product on the pack can be maintained during processing.

Sweetening agents, flavouring agents and other taste components can be selected during formulation development to provide a known balanced sensory experience without affecting the other ingredients.

Production of a Multi-Component Iron, Calcium, Vitamins and Minerals Liquid

The liquid can be prepared by addition of all ingredients to purified water in a sequential manner. The time, temperature, speed of addition and order of addition can be established during process development and then transferred to the manufacturing process.

Bulk samples can be taken during this step and analysed for colour, clarity, pH, viscosity, temperature and other parameters. In-process homogeneity can be monitored and the product can be tested for key ingredients before formulation filling is initiated.

Assay of vitamins, minerals, iron, calcium, the cyanocobalamin component and antioxidant elements can be performed by the relevant analytical methods. Microbiological quality control can also be evaluated.

Stability of a Multi-Ingredient Range for Pharmaceutical Development

Samples of a formulated product can be tested for a range of physical parameters such as pH, appearance, colour, viscosity, presence of sediments, clouding or flocculation, suspension stability and sedimentation rate. Assays for all ingredients can be performed as indicated to monitor potential deterioration.

Additional testing can be performed for microbial growth, light- or heat-induced degradation, partial or total precipitation or change in appearance.

The product can be stored within glass bottles made from suitable materials and bottle compatibility can also be evaluated.

Final Packaging Preparation of an Iron, Calcium, Vitamins and Antioxidant Syrup

Samples of a formulated range can be filled into pharmaceutical-grade bottles under a controlled filling environment. Final pack can be examined for appropriate fill volume, closure fit, leak resistance, and cleanliness. The product can also be evaluated during storage over time for the same stability parameters as already discussed. Chemical, physical, and microbiological parameters can be monitored as required.

This final step completes the overall PCD Pharma Franchise Product development process of the multi-ingredient oral liquid formulation and can support systematic product development for this type of multi-active range.
