KETOCONAZOLE 2%W/V + ZINC PYRITHIONE 1%W/W SHAMPOO

Ketoconazole 2% W/V + Zinc Pyrithione 1% W/W Shampoo This is a co-formulated medicinal topical cleansing composition developed by combining the two active ingredients in a suitably formulated shampoo base. Ensuring the even dispersal of both active ingredients and other components is often part of formulation development as their physical properties must be maintained in line with the selected surfactants and formulating excipients. For formulators looking to expand their dermatology catalog, this product can also be included as part of a PCD Pharma Franchise line.

The composition contains: Ketoconazole 2% W/W and Zinc parathion 1% W/W. Other ingredients of the shampoo base may include surfactants, viscosity adjusting agents, stabilizers, preservatives, hair conditioning agents, flavoring agents and other formulating excipients as approved by the product design. Compatibility, physical stability and finished-product quality are all taken into consideration when designing the formulation.

Formulation Composition and Ingredient Compatibility

Developing a batch of Ketoconazole 2% W/V + Zinc Parathion 1% W/W Shampoo begins with selection of a shampoo base that can maintain the required physical properties throughout its shelf life. Surfactants may be important ingredients in the base and are selected according to the cleansing behavior needed and their compatibility with the active ingredients used.

Because Zinc Parathion is typically handled as a dispersed ingredient in the formulation, attention should be paid to particle dispersal and suspension properties. Incorporation techniques can facilitate uniform dispersal of both active ingredients to ensure a consistent finished shampoo. Other excipients might include humectants, viscosity-raising agents, preservatives, chelators, hair conditioners and scent ingredients. The quantities of these excipients will depend on the approved product composition.

Active ingredient compatibility studies can highlight any potential issues that may arise between Zinc Parathion and the shampoo base before preparing manufacturing lots. Dispersed Zinc Parathion requires particular care to ensure appropriate dispersal in the main lot while the physical form can influence the batch-formation procedure.

Manufacturing and dispersing the active ingredients can require specific mixing time and speed guidelines, depending on the physical properties of the batch. Dispersion stability can be closely monitored and could require deaeration or homogenization steps, as needed, to achieve suitable finished-product qualities.

After dispersing the active ingredients, quality control checks can be carried out before the additional preparation of the bulk product. It is then ready for filling operations after quality evaluations have been completed.

Physical Property Control and Quality Assurance

Maintaining physically stable compositions during manufacturing processes is an important consideration when formulating Ketoconazole 2% W/V + Zinc Parathion 1% W/W Shampoo. Viscosity and color are two such parameters that can be monitored from batch to batch and could vary depending on processing conditions. Unacceptable variation in viscosity can interfere with packaging and product handling.

PH can also be a focus of process control to prevent pH fluctuations or variations in dispensed product from reaching the customer. Product appearance, color, odor, uniformity, sedimentation and dispersal of the dispersed Zinc Pyrithione might be assessed during manufacturing, along with batch weight and viscosity measures.

Analytical testing of the active ingredients can be performed to confirm the disbursement is appropriate. Viscosity and appearance testing can be conducted on a finished product batch prior to filling.

Process stability can be evaluated by storage at selected conditions, including monitoring appearance, color, viscosity, pH, Zinc Parathion dispersal and other parameters of interest. Unacceptable variation can be used to define the suitable storage conditions and shelf life. Packaging material can also influence the physical stability of the finished product and should be considered during development.

Other Physical Parameter Evaluation

pH can be controlled to a predetermined acceptable range and viscosity can be maintained within limits during manufacturing by process control. Homogeneity of the finished shampoo can be assessed regularly. Sedimentation behavior of Zinc Pyrithione particles can be a factor to consider, especially if the aim is to ensure the dispersal is uniform for use by the end user.

In-process assessment of both time and batch characteristics can include weight, viscosity, pH, appearance and other parameters according to the quality standard. Each individual physical characteristic can influence the quality of the finished shampoo and whether it can be successfully stored and used over its intended shelf life.

Analytical and Finished Product Testing

Finished-product quality-testing can include verification of the identity and potency of the two active ingredients. Additionally, appearance, color, viscosity, final batch weight or volume, homogeneity and any other relevant physical properties can be checked. Microbiological tests may also be performed for acceptability of the finished product.

These tests can supplement the physical and chemical evaluations and ensure the product can be stored without risk of microbial spoilage. Microbiological test results may be obtained for the finished product to supplement physical evaluations.

Product stability studies can support the shelf-life assessment by evaluating potential change in physical characteristics during storage at chosen conditions. Appearance, color, viscosity, odor and other specifications can be monitored. These observations can inform setting of the product shelf-life, the appropriate storage conditions, the storage packaging and the suitability for use until the date specified.

Particle- dispersion stability can be an aspect of stability evaluation. Maintaining an appropriate dispersal of Zinc Pyrithione particles throughout their storage life can be a challenge with some formulations.

Packaging Development and Broader PCD Franchise Portfolio

A selection of packaging materials can be evaluated for inclusion in the manufacturing process to best protect the finished products from the environment during storage and transport. Items to evaluate may include bottle material, closure type, filling performance and compatibility with the formulation. Completed bottles and labels should be prepared with the proper product-identification and safety and usage information. When maintaining a consistent approach to batch manufacture and the finished-product specification, a dermatologist in marketing can use this formulation as part of a broader PCD portfolio of topicals.
